海参肠道β-1,3-葡聚糖酶的提取条件及其酶学性质 被引量:8

摘要 对海参肠道β-1,3-葡聚糖酶的提取条件及酶学性质进行了研究.结果表明,最佳提取条件为:pH6.0的磷酸盐浸提液0.05mol/L,NaCl浓度为0.05mol/L,缓冲液体积与海参肠质量之比为4:1,硫酸铵饱和度为80%;该酶最适反应条件为:温度40℃,pH6.0;该酶在10-40℃,pH5.O~6.5酶活力稳定.Mn^2+对酶具有一定的激活作用,Cu^2+、Cd^2+、Mg^2+、Fe^2+、Zn^2+、Bd^2+、K^+、Ag^+对酶存在不同程度的抑制,其中Cu^2+对酶的抑制作用最强. Extraction and properties ofβ-1,3-glucanase from the gut of sea cucumber were investigated in this paper. The optimum extraction condition was obtained as follows, phosphate buffer (pH 6.0, 0.05 mol/L, 0.05 mol/L NaCl)- gut=4 : 1,saturation of (NH4)2SO4 was 80% The optimum temperature and pH of the enzyme were 40 ℃ and pH 6. 0. The crude β-1,3-glucanase was stable at pH 5.0-6.5. The enzyme was activated by 0.05 mol/L Mn^2+ and inhibited by Cu^2+ , Ca^2+ , Mn^2+ , Mg^2+ , Fe^2+ , Zn^2+ , Ba^2+ , K^+ and Ag^+ , in which Cu^2+ was strongest inhibitor.
