柱切换HPLC法测定法莫替丁血药浓度及其在药动学研究中的应用 被引量:2

摘要 目的:建立简便有效、适用于药动学研究的法莫替丁血药浓度高效液相(HPLC)测定方法。方法:采用柱切换HPLC技术。分析柱(150mm×4.6mm,ID)固定相为YWGC18(5μm),分析流动相为甲醇与乙酸铵缓冲液;预处理柱(50mm×4.6mm,ID)填料为YWGC18(10μm),预处理流动相为乙酸铵缓冲液。结果:法莫替丁血浆浓度标准曲线线性范围为5~200ng/ml(r=0.9998),血浆中最低检测质量浓度为4ng/ml。方法日内及日间精密度分别为2.97%±1.56%,6.04%±1.62%,回收率为98.37%±2.91%。结论:应用本方法研究法莫替丁在正常人体内的药动学,具有简便迅速、准确灵敏、易于实现自动化等特点。 Objective:To establish a simple and effective HPLC method for determination of famotidine in plasma for pharmacokinetic study. Methods: A HPLC method with column switching technique was used. The analytical column(150 mm×4.6 mm,ID)was packed with YWG C 18 (5 μm)and the pretreating column(50 mm×4.6 mm,ID) was packed with YWG C 18 (10 μm); A solution of 0.2 mol/L ammonium acetate (adjusted pH to 3.04 with sulfuric acid) was used as pretreating mobile phase and 14%(V/V) methol in it was used as analytical mobile phase. The wave length was 266 nm. Results: The method was linear with the plasma concentration range of 5 ~200 ng/ml(r=0.9998), and the detection limit was 4 ng/ml.The precision was 2.97%±1.56% for within day and 6.04%±1.62% for between days respectively. The relative recovery of the method was 98.37%±2.91% . [WT5HZ]Conclusion:The method is easily to be applicated automatedly as it is simple, rapid, accurate and sensitive.
