白藜芦醇通过调节肾素-血管紧张素系统抑制白血病细胞增殖 被引量:2

Resveratrol Inhibits Proliferation of Leukemia Cells by Regulating Renin-Angiotensin System

摘要 目的观察急性白血病患者骨髓中肾素活性,探讨白藜芦醇对白血病U937细胞增殖抑制作用。方法采集32例急性白血病患者初诊时的骨髓和外周血,以放射免疫法检测标本中的肾素活性,9例缺铁性贫血患者作为对照。采用一定浓度梯度(25~200μmol/L)白藜芦醇作用于人白血病U937细胞,培养不同时间(12、24、36、48h)后,以MTT法检测U937细胞增殖抑制率。选择100、200μmol/L白藜芦醇作用U937细胞,培养48h后,采用放射免疫法检测细胞培养上清液中血管紧张素Ⅱ含量。结果急性白血病患者骨髓组织中肾素活性高于对照组骨髓及自身外周血中的肾素活性,两两相比差异有统计学意义。白藜芦醇作用U937细胞后,培养细胞上清液中血管紧张素Ⅱ含量明显降低。白藜芦醇呈时间与浓度依赖性抑制白血病U937细胞增殖。结论白藜芦醇可能通过调节肾素-血管紧张素系统,下调血管紧张素Ⅱ的生成而抑制白血病细胞增殖。 Objective To study the renin activity of bone marrow from de novo patients with acute leukemia, and to explore the inhibitory effects of resveratrol on proliferation of human leukemia U937 cells. Methods Human bone marrow and peripheral blood samples were collected from 32 de novo patients with acute leukemia and 9 patients with iron deficiency anemia serving as controls. The renin activity was examined by radioimmunoassay. Human leukemia U937 cells were treated with different concentrations of resveratrol (25-200 μmol/L) for different time periods (12, 24, 36, 48 h). The cell proliferation was determined by MTT assay. The content of angiotensin Ⅱ was detected by radioimmunoassay. Results The renin activity of bone marrow in patients with acute leukemia was higher than that of peripheral blood in those patients and that of bone marrow in controls. Resveratrol down-regulated the secretion of angiotensin Ⅱ in U937 cells. Resveratrol inhibited proliferation of U937 cells in a dose- and time-dependent manner. Conclusion Resveratrol inhibits proliferation by regulating renin-angiotensin system to down-regulate the secretion of angiotensin Ⅱ in leukemia cells.
