骨疏丹指纹图谱分析及其活性相关性研究 被引量:3

摘要 目的建立骨疏丹的高效液相色谱指纹图谱,分析其成分与促成骨样细胞增殖活性的相关性,确定活性成分并推荐质量控制指标。方法色谱条件为:色谱柱为DiamonsilTMC18,流动相为20 mmoL.L-1NaH2PO4溶液和乙腈,采用梯度洗脱,柱温为25℃,检测波长为270 nm。对用撤药法拆方所得处方进行HPLC色谱指纹图谱分析和促成骨样细胞的增殖活性试验,采用相关分析和回归分析将活性数据分别与各指纹峰峰面积及组方药味相关联。结果指纹图谱分析方法的精密度、重复性、稳定性均符合有关规定。从指纹图谱中得到了18个主要色谱峰,其中9个色谱峰与活性相关,选择淫羊藿苷、柚皮苷作为质量控制指标。配伍关系研究结果与中医组方原则一致。结论骨疏丹指纹图谱可为骨疏丹的质量控制和活性及配伍关系研究提供了依据。 Objective To study the HPLC fingerprint of Gushudan (traditional Chinese medicines) prescription, correlate the constituents with osteoblastic proliferative activity and find quality control indices. Methods The chromatographic conditions were as follows: Diamonsil^TM C18(5μ, 200 mm × 4.6 mm) column; mobile phase: 20 mmoL· L^-1 NaH2PO4 and CH3CN, gradient elution; column temperature: 25℃ ; flow rate: 1.0 mL/min; detection wavelength: 270 nm. Gushudan prescription was disassembled into 11 recipes according to the design of decomposed recipes. The fingerprint ehromatograms of the recipes were completed by the HPLC method and the proliferative activity of osteoblast-like cells was assayed for each recipe. Regression and correlation analysis were applied to correlate the area of each peak and the activity data. Results There were 18 peaks in the fingerprint chromatogram of Gushudan prescription. The RSDs of precision, stability and repeatability were less than 3 %. Nine peaks were correlated with the proliferative activity. Icariin from Herba Epimedii and naringin from Rhizoma Drynariae were selected as the quality control indices of Gushudan. Conclusions This study can be the base of the quality control and the investigation of compatibility for Gushudan prescription.
