摘要
[目的]探讨白血病抑制因子(leukemia inhibitory factor,LIF)在骨性关节炎(OA)关节组织中的表达规律及临床相关性。[方法]切取(32人)73例骨性关节炎滑膜、软骨和软骨下骨组织,含10人正常关节组织标本,应用RT-PCR检测LIF mRNA在OA不同病变阶段中的表达,酶联免疫吸附反应(ELISA)检测LIF在组织中的含量。[结果]中度OA病变软骨下骨LIF mRNA表达量最高。重度OA软骨中LIF mRNA的表达量最高,软骨下骨的表达量最低。ELISA检测LIF在各组织的含量情况与上述结果一致。[结论]LIF在OA组织中表达呈现出一定的时间及空间分布的规律,提示LIF作为一种分解代谢因子可能参与了OA发病过程。
[Objective] To investigate the regulation expression of leukemia inhibitory factor(LIF) in human normal and osteoarthritic(OA) articular tissue and related clinical changes. [Methods] 73 samples of human articular tissues were obtained from operational procedures and the good quality RNA were isolated using Magnetic Beads. These samples were donored by patients who underwent articular operations. Using the reverse transcription-polymerase chain reaction(RT-PCR), the mRNA express of LIF was measured by semi-quality and the location of LIF protein was determined by enzyme-linked immunosorbent assay(ELISA). [Results] LIF mRNA was expressed to very low level in normal articular tissues and there was no significant difference in different anatomical locations. In moderate degrading subchondral bone, LIF mRNA was expressed to its highest level. LIF was expressed to the highest level in seriously degrading articular cartilage tissue. This result was similar with ELISA testing results. [Conclusions] The different temporal and special distributions of LIF in normal and OA articular tissues imply that LIF may play some important roles in the pathogenesis of OA.
