摘要 目的研究颈椎后路减压手术对颈椎后纵韧带中骨形态,发生蛋白(Bone Morchopedic Protein,BMP)-4mRNA表达的影响。方法选用SD大鼠72只,随机分为三组。一组为手术组行颈椎全椎板切除减压加椎旁肌切断手术(手术节段C3-C6);二组为假手术组,仅作皮肤切口;三组不作任何处理为空白组。按手术后不同时间点(1、2、4、8周)处死动物,取其后纵韧带。采用半定量RT-PCR方法检测组织BMP-4 mRNA表达水平。结果空白组及对照组未见明显BMP-4 mRNA的表达;手术组于手术后1周表达明显,持续到8周。结论颈椎后路减压手术可以导致后纵韧带早期细胞BMP-4mRNA表达上升,内源性的BMP-4可能导致手术后韧带骨化发展。 Objective: To study the effect of BMP-4 in the course of Posterior Longitudinal ligament ossification through gene analysis. Methods : The tissue of posterior longitudinal ligament of experimental segment was resected in 1,2, 4 and 8 weeks after operation form laminectomy and muscle cutting on C3-6 level ( group 1 ). The total RNA was distracted from the pathological tissue. The expression degree of BMP - 4 mRNA was detected using methods of RT-PCR. The group of exposure was set up only as experimental control group (EC group). Animals without operation was in negative control group ( group 3). Results : There wssnt significant difference betweeen the group B and group C. The expression of BMP - 4 mRNA was the highest in the tissues resected from the animals 1 weeks after operation in group A. This expression continued in the 8th weeks. The changes of expresion degree were related positively to the cervical opration. BMP-4 mRNA increased significantly in the 1st group compared with the contrasting group. Conclusion: The operation of posterior decom- pression can lead to the increase in the express of BMP -4 mRNA, which can probably cause OPLL.
