脂质体转染前庭椭圆囊上皮细胞的影响因素

摘要 利用阳离子脂质体-Lipofectamine TM2000介导真核表达质粒（pRK5·EGFP）转染椭圆囊上皮细胞（UEC）。当UEC培养至60％-100％融合时，用不同脂质体／DNA，分别转染6～24h，发现UEC融合80％-90％，DNA（μg）／脂质体（μl）=1：3，转染8h时转染效率最高。认为LipofectamineTM2000可有效的介导真核表达质粒转染UEC，其转染效率与细胞生长状态、脂质体／DNA、转染时间直接相关。
