摘要 Aurora蛋白激酶家族,包括Au-rora-A、Aurora-B和Aurora-C,在细胞有丝分裂期过程中发挥着重要作用。Aurora-A定位于中心体和纺锤体,主要参与中心体的成熟、分离和纺锤体形成,还参与p53通路、细胞凋亡和有丝分裂的调节等。Auro-ra-A在多种肿瘤组织中存在着高表达。它的高表达可能会导致基因的不稳定和肿瘤的发生,与某些肿瘤的预后相关。最近对其三维结构的测定推进了对激酶抑制剂的研究,希望能够发现新的抗癌因子。Aurora-B参与染色体调节、分离,并在纺锤体检查点等方面发挥重要作用。Auro-ra-C和Aurora-B共同参与哺乳动物有丝分裂染色体分离和胞质分裂。 Aurora kinases include Aurora A, Aurora B anti Aurora C. They play a crucial role in cellular division. Aurora A gene is located within mitotic spindle and centrosome. It participates in centrosome maturation and segregation, spindle assembly and cytokinesis, and also, it participates in the p53 pathway,apoptosis and the regulation of mitosis. The research demonstrates Aurora A is over-expressed in diverse solid tumors. Aurora A overexpression contributes to genetic instability and tumorigenesis, and it has some relationship with the prognosis of some tumors. The rencent determination of the three-dimensional structure of Aurora A has boosted the search and development of inhibitors of Aurora kinases, which might also function as novel antioncogenic agents. Auro- ra-B participates in chromosome alignment and segregation, and plays an important role in spindle checkpoint. Aurora-C cooperates with Aurora-B to regulate mitotic chromosome segregation and cytokinesis in mammalian cells.
