摘要
从鹰嘴豆种子中分离到3种β-半乳糖苷酶(酶Ⅰ、酶Ⅱ和酶Ⅲ)。将酶Ⅰ和酶Ⅱ纯化后,其比活力分别提高了19倍和48倍,测得它们的表观分子量分别为24000和58000。酶Ⅰ含18种氨基酸,171个氨基酸残基;酶Ⅱ含16种氨基酸,305个氨基酸残基;
Three Kinds of β gatactosidase(Ⅰ、ⅡandⅢ)were separated from gram chicken bean by ammonium sulphate fractionation(30%~70%saturation)and by ion—exchange chromatography on DEAE—Cellulose—32.β Galactosidase ⅠandⅡwere purified by subsequent chromatographic separation on CM—cellulose—52.Their specific activities were improved by 19 times and 48 times respectively.The apparent molecular weights(Mr)of the two enzymes were 24000 and 58000 respectively.The amino acid composition of β galactosidase Ⅰ consists of 18 Kinds of amino acids and 171 amino acid residues,While that of β galactosidase Ⅱ consists of 16 Kinds of amino acids and 305 amino acid residues and the content of Histidine and Proline in the latter is zero.
