摘要
目的:观察不同浓度的苦参碱对宫颈癌Hela细胞的作用。方法:浓度分别为0.5mg/ml、1.0mg/ml、1.5mg/ml、2.0mg/ml的苦参碱体外培养Hela细胞24h、48h、72h后,分别用MTT法观察苦参碱对Hela细胞的生长抑制作用、AnnexinV-PI双标染色用流式细胞仪检测苦参碱对Hela细胞凋亡的影响。结果:不同浓度的苦参碱具有抑制宫颈癌Hela细胞增殖的作用,呈明显的时间、剂量依赖性。各浓度和时间之间比较具有显著差异性(P<0.01)。FCM检测结果显示苦参碱作用后,细胞的凋亡率增加,呈现随时间和剂量依赖性。各浓度和时间之间比较也具有显著差异性(P<0.01)。结论:苦参碱对人宫颈癌Hela细胞的增殖具有明显的抑制作用,能诱导肿瘤细胞的凋亡。
Objective: To study the effect of Matrine with different concentrations on cervical cancer cell. Methods Matrine with different concentrations (0. 5mg/ml, 1.0mg/ml, 1. 5mg/ml, 2. 0mg/ml) was used to cultive cervical cancer Hela cells in vitro. After Hela cells cultived 24h,48h and 72h respectively,observe the effect of Matrine on cervical cancer cell was measured with MTT means and flowcytometry(FCM). Results: Matrine inhibited the proliferation and induced apoptosis of Hela cell on dose and time-depending manner, and the differences among concentrations and times were significant(P〈0. 01). Conclusion: Matrine restrained the prolifiration of Hela cell significantly,also it can induce the apoptosis of cervical cancer.
