摘要
目的探讨基质金属蛋白酶-9(MMP-9)、血管内皮细胞生长因子(VEGF)的表达与肾细胞癌血管生成及生物学特性关系。方法应用免疫组化SP法检测78例肾癌MMP-9、VEGF的表达及微血管密度(MVD)。结果MMP-9在浸润进展期肾癌组(T3~T4)的阳性表达率为87%,明显高于局限性肾癌组(T1~T2)的50%(P〈0.01);MMP-9和VEGF同时表达阳性者,其血管密度为140.86±24.87,明显高于仅MMP-9或VEGF表达阳性的:99.3±47.97(P〈0.01),亦明显高于二者都表达阴性的:80.16±33.83(P〈0.0001);肾癌MMP-9、VEGF的表达呈显著性正相关(r=0.40966,P〈0.001),MMP-9、VEGF在肾癌的血管生成中有协同作用。结论MMP-9、VEGF联合检出有助于预测肾癌侵袭和转移。
Objective To study the expressions of matrix metalloproteinase-9 (MMP-9), vascular endothelial growth factor (VEGF) and their correlation with angiogenesis and biological behaviour in renal cell carcinoma(RCC). Methods The expressions of MMP-9 ,VEGF and CD34 were detected by means of immunohistochemistry in 78 cases of RCC. Results The positive expression rate of MMP-9 was 87% in the high staging group and 50% in the low staging group,the statistical significance existed between the two groups (P〈0.01) ;the MVD was 140.86±24.87 in both positive expressions of MMP-9 and VEGF, was higher than that in the single positive ones(99.3±47.97)(P±0.01)and the both negative ones (80. 16±33.83)(P〈0. 000 1). The expression of MMP-9 correlated with VEGF in renal cell carcinoma(r=0. 409 66, P〈O. 01). Conclusion MMP-9 and VEGF may serve as biological predictor for tumor metastatic potential in renal cell carcinoma.
