氟西汀对慢性应激大鼠行为及心肌ICAM-1和VCAM-1表达的影响 被引量:2

摘要 目的研究氟西汀对慢性应激大鼠行为及心肌细胞间粘附分子-1(ICAM-1)、血管细胞粘附分子-1(VCAM-1)表达的影响。方法将青年(8周龄)雄性Wistar大鼠24只随机分为对照组、应激组、氟西汀组各8只。应激组和氟西汀组大鼠单笼饲养,实验第1~21天,接受各种不同的应激。氟西汀组大鼠每天给予氟西汀水溶液灌胃。对照组大鼠群养不给任何刺激。实验第22天测量大鼠的体质量及用Open-field法测量大鼠的行为,然后杀死所有大鼠,速取左心室心尖部心肌组织,制成免疫组化切片,免疫组化法检测蛋白ICAM-1、VCAM-1表达,进行图像分析。结果与对照组比较,应激组大鼠体质量明显下降,水平穿越格数、直立次数、修饰次数减少,粪便粒数、中央格停留时间增加。与应激组大鼠比较,氟西汀组大鼠体质量明显增加,水平穿越格数、直立次数、修饰次数增加,粪便粒数、中央格停留时间减少。应激组大鼠心肌ICAM-1表达高于对照组[OD值分别为(23.89±10.19),(42.44±12.36),t=3.27,P〈0.01]、VCAM-1表达高于对照组[OD值分别为(8.29±2.01),(20.61±7.11),t=4.71,P〈0.01];氟西汀组大鼠心肌ICAM-1表达低于应激组[OD值分别为(42.44±12.36),(29.86±5.97),t=2.59,P〈0.05]、VCAM-1表达低于应激组[OD值分别为(20.61±7.11),(13.11±2.11),t=2.86,P〈0.05]。结论慢性不可预见性应激可引起大鼠体质量下降及行为学改变,引起大鼠心肌炎症性反应。应用氟西汀可以预防慢性应激引起的大鼠体质量下降及行为学改变,减轻心肌炎症性反应。 Objective To study the effects of fluoxetine on the behavior and the expression of intercellular adhesion molecule-1 ( ICAM-1 ), vascular cell adhesion molecule-1 ( VCAM-1 ) in myocardium in chronic stress rats. Methods Male Wistar rats were divided into control group, stress group and fluoxetine group randomly, 8 rats every group. Stress group and fluoxetine group were fed separately one rat in one box. From 1 -21 days,rats were applied various types of stresses. At the same time, fluoxetine group were given fluoxetine every day. The control group were fed in two boxes,and no stress from 1 -21days. On 22th day,the change of the behavior of the rats were observed and killed all the rats. Two small pieces of myocardium were immediately dissected from cardiac apex of left ventricles respectively. Sections for immunohistochemistry were made. Used immunohistochemistry method to measure the expression of ICAM-1/VCAM-1 protein in myocardium and analyze optical density (OD) average of ICAM-1/VCAM-1. Results After 21 days stress, weight, ambulation, rearing and grooming decreased and stopping time in center, defecation increased in stress group. Compared with stress group, weight, ambulation, rearing and grooming increased and stopping time in center, defecation decreased in fluoxetine group. ICAM- 1/VCAM-1 expressed obviously in stress group , and the expression of ICAM-1 ( 23.89 ± 10.19,42.44 ± 12.36, t = 3.27, P〈0.01),VCAM-1 (8.29 ±2.01,20.61 ±7.11, t=4.71, P〈0.01) in myocardium in rats of stress group were higher than those in fluoxetine group. The expression of ICAM-1 (42.44 ± 12.36,29.86 ± 5.97, t = 2. 59, P〈0.05) ,VCAM-1 (20.61±7.11,13.11 ±2.11, t=2.86, P〈0.05) in myocardium in rats of stress group were higher than those in fluoxetine group. Conclusions Chronic stress result in weight decrease and the behavior changes of rats. Chronic stress induced overexpression of ICAM-1 ,VCAM-1 in myocardium . Fluoxetine can prevent weight decrease, behavioral change and the overexpression of ICAM-1, VCAM-1 in myocardium in chronic stress rats.
