心理应激性失眠大鼠中缝背核神经元超微结构及加味逍遥散的影响 被引量:5

摘要 目的观察心理应激性失眠大鼠中缝背核神经元超微结构变化,探讨加味逍遥散治疗对其的影响。方法30只雄性Wistar大鼠随机分为对照组、模型组和药物组。采用限制性制动方法复制心理应激性失眠模型,药物组给予加味逍遥散连续灌胃15d。多导睡眠描记观察睡眠-觉醒指标变化情况。灌注固定后应用透射电镜观察各组中缝背核神经元超微结构。结果与对照组比较,模型组大鼠造模后觉醒时间增加(第3天时,对照组33.44%,模型组44.02%,P〈0.01;第15天时,对照组36.47%,模型组41.25%,P〈0.05)、非快速眼动(NREM)睡眠时间减少(第3天时,对照组55.64%,模型组44.02%,P〈0.01;第15天时,对照组55.89%,模型组50.10%,P〈0.05);经加味逍遥散治疗后得到恢复。电镜下可见心理应激性失眠大鼠中缝背核神经元细胞核染色质浓缩并边集;粗面内质网扩张、排列紊乱和脱颗粒;线粒体肿胀,部分空泡样变性;高尔基复合体肿胀和极性消失;髓鞘板层排列疏松、间隙加大,内部轴突神经丝松解、弯曲;突触结构分界不清,突触小泡稀疏。药物组与对照组比较无明显病变。结论应激性失眠时,中缝背核神经元出现一系列超微病理改变,经加味逍遥散治疗后超微结构得到好转。 Objective To investigate the uhrastructural changes in the nucleus raphes dorsalis neurons of psychological stress insomnia rats and further to observe the therapeutic effects of Jia Wei Xiao Yao San. Methods Thirty male Wistar rats were divided randomly into control group, model group, and experimental group. The psychological stress insomnia rat model was induced by restricting activity. Jia Wei Xiao Yao San was given by lavaging for 15 days. Sleep-waking cycle was observed by polysomnogram. The uhrastructures of raphe nuclei neurons were observed with transmission electron microscope (TEM). Results Compared with the control, the ratio of wake increased (on day 3, 33.44% in control group vs 44.02% in model, P〈 0.01; on day 15,36.47% in control group vs 41.25% in model, P〈0.05) and the ratio of non-rapid eye alovement( NREM)sleep decreased (on day 3,55.64% in control group vs 44.02% in model, P〈0.01 ; on day 15,55.89% in control group vs 50.10% in model, P 〈 0.05 ) after restriction, which were improved after therapy of Jia Wei Xiao Yao San, Compared with control group, most model group raphe nuclei dorsalis neuron's nucleus showed chromatin condensation and margination, rough endoplasmic reticulum (RER) swelling, derangement, and ribosomal granules broken. In addition, most mitochondria exhibited swelling and vacuole degeneration, as well as Golgi's complex exhibited swelling and polarity disappearance. Moreover, myelin sheath broke up and synapses degenerated. The uhrastructures of the raphe nuclei' s neuron of experimental group were similarly with those of the control group. Conclusion Lots of ultrastructural changes in raphe nuclei dorsalis neurons induced by psychological stress insomnia and treatment by Jia Wei Xiao Yao San could provide morphological evidences for further investigation of association between raphe nuclei and psychological stress insomnia with depressive disorder and the therapeutical effect of Jia Wei Xiao Yao San.
