摘要
目的:检测裙带菜多糖(UPPS)对人肝癌HepG-2细胞生长的抑制作用,并探讨其诱导细胞凋亡和抗肿瘤作用的关系。方法:采用MTT法、流式细胞术检测UPPS对HepG-2细胞生长的抑制作用和诱导细胞凋亡的作用;采用免疫细胞化学染色,观察Bax和Bcl-2表达水平的变化。结果:UPPS能抑制HepG-2细胞增殖,明显高于对照组(P<0.01)。诱导细胞凋亡作用发生在UPPS处理24~48h;UPPS处理HepG-2细胞后,Bax蛋白表达增强,Bcl-2蛋白表达下调。结论:UPPS对HepG-2的抑制作用是通过诱导细胞凋亡实现的,可能与Bcl-2和Bax表达水平的变化有关。
Objective: To investigate the inhibitory effect of Undaria pinnatifida polysaccharides (UPPS) on hepatocellular carcinoma HepG-2 cells and its possible mechanism. Method: The effect of inhibiting proliferation and inducting apoptosis of UPPS were determined by means of MTT and FCM. The expression of Bcl-2 and Bax proteins was immunohis to chemcally evaluated after treatment of UPPS. Results: UPPS inhibited HepG-2 cells growth in vitro, significantly higher than the negative control group (P〈 0.01), and induced apoptosis after UPPS treatment 24~48h, with decreased expression of Bcl-2 and increased expression of Bax. Conclusion: UPPS inhibits the proliferation of HepG-2 cells by apoptosis, perhaps associated with the change of expression of Bax and Bcl-2.
