鲁米诺-H_2O_2体系流动注射-化学发光增强法测定胰蛋白酶 被引量:2

Flow Injection Analysis of Trypsin with Enhanced Chemiluminescence Detection by the System Luminol and Hydrogen Peroxide

摘要 在碱性条件下,胰蛋白酶对鲁米诺-过氧化氢体系的化学发光体系具有显著的增强作用,据此建立了流动注射-化学发光增强法测定胰蛋白酶的新方法.该法简单、快速、线性范围宽,在优化的实验条件下,测定胰蛋白酶在1.0~50.0mgL/的浓度范围内相对发光强度与其浓度呈良好的线性关系,检出限可达0.26mg/L,对浓度为10.0mg/L的胰蛋白酶进行平行测定其相对标准偏差(RSD)为0.7%(n=11),成功地用于合成样品中胰蛋白酶的测定,回收率在90.1%~104.1%,结果令人满意。 It was found that in an alkaline medium, the chemiluminescence(CL) intensity of the reaction between hydrogen peroxide and luminol was greatly enhanced at the presence of trypsin. Based on the fact, a new CL method for the determination of trypsin was proposed in combination with the flow injection (FI) technique. The method is simple, rapid and effective to determine trypsin in the range of 1.0 - 50.0 mg/L with a detection limit of 0.26 mg/L. The relative deviation is 0.7% for the determination of 10.0 mg/L trypsin( n = 11). The method has been applied to determine the concentration of trypsin in synthetic samples with satisfactory results.
