摘要
目的:探讨高脂饲料加剧阿霉素(ADR)肾病大鼠高脂血症及肾组织形态学改变的作用。方法:用ADR静脉注射建立大鼠肾病模型,饲喂含15%胆固醇、5%猪油的高脂饲料。观察大鼠尿蛋白排泄量,测定血清中脂质有关指标以及检查肾组织形态学改变。结果:饲喂高脂饲料的ADR肾病大鼠,血清脂质水平在整个试验过程中均明显高于饲喂基础饲料的ADR肾病大鼠,肾小球病变也显著加重。
OBJECTIVE Abnormal lipid metabolism is a frequent complication in renal diseases and plays an important role in the progression of renal injury. Adriamycin(ADR) induced nephritis was used to evaluate the effects of hyperlipidemic chow on the progression of renal damages in this study. METHODOLOGY Forty male Wistar rats were used in this study. ADR(6 0 mg/kg) was used for induction of nephritis in 20 rats. 10 nephritic rats and 10 normal rats were fed with hyperlipidemic chow(1 5% cholesterol, 0 5% cholic acid and 5% pig tallow added to the basic rodent chow) for 13 weeks, while 10 nephritic rats and 10 normal rats with basic rodent chow. Urinary protein excretion, plasma lipids levels were investigated and renal histologic changes were observed after 13 weeks. RESULTS At the 4, 8 and the 13th week of observation, proteinuria in nephritic rats fed with hyperlipidemic chow increased more significantly than in nephritic rats fed with basic chow(15 91±5 74 vs 11 05±8 97 mg/24h at the 4th week, 20 51±8 53 vs 13 23±3 91 at the 8th week, 29 73±7 94 vs 20 94±6 28 mg/24h at the 13th week, P <0 01); while in the normal rats fed with hyperlipidemic chow, proteinuria also increased significantly in the 8th and 13th week(4 22±2 58 vs 1 99±1 69mg/24h, P <0 05; 12 11±4 69 vs 1 98±1 89 mg/24h, P <0 01). In nephritic rats fed with hyperlipidemic chow, serum total cholesterol, tryglyceride, low density lipoprotein cholesterol and very low density lipoprotein cholesterol increased significantly than those in nephritic rats fed with basic chow at the 13 week of observation( P <0 05 for all); while between normal rats fed with hyperlipidemic chow and basic chow, mild elevations of total cholesterol, low density lipoprotein cholesterol and very low density lipoprotein cholesterol were found. In the histologic study of the renal damages, exacerbated glomerular changes and increased foam cells were found in the nephritic rats fed with hyperlipidemic chow as compared to those in nephritic rats fed with basic chow. CONCLUSION Hyperlipidemic chow increases proteinuria and lipids levels in ADR induced nephritic rats and exacerbates the renal damage through a mechanism in which hyperlipidemia plays an important role.
