摘要
目的 观察参附注射液对大鼠肢体缺血再灌注后骨骼肌组织髓过氧化物酶(MPO)、丙二醛(MDA)含量和细胞色素氧化酶(CCO)及钙激活ATP酶(Ca^2+-ATPaSe)活性水平变化的影响,探讨其作用机制。方法 SD大鼠30只,建立肢体缺血再灌注损伤模型。A组:对照组(10只),只做手术,不做阻断;B组:缺血再灌注组(10只),缺血4h,再灌注4h,未作药物治疗;C组:参附注射液治疗组(10只),缺血及再灌注时间同前一组。阻断前于股动脉阻断部位处注入参附注射液(2.0mL/kg),5min后夹闭股动静脉。再灌注前15min,再次自尾静脉注入同样浓度参附注射液,检测骨骼肌组织MPO、MDA、湿重/干重值(W/D)和CCO及Ca^2+-ATPase活性的变化。结果 参附注射液能降低大鼠骨骼肌缺血再灌注损伤所引起的骨骼肌组织MPO、MDA、W/D的升高,在一定程度上保护CCO及Ca^2+-ATPase的活性。结论 参附注射液可减轻肢体缺血再灌注时骨骼肌组织的损伤。
Objective To observe the effect of shenfu injection on ischemia/reperfusion injury in skeletal muscle of rats. Methods Rat posterior limb ischemia/reperfusion model were made. Thirty SD rats were randomly divided into three groups (n =10): sham group (group A), ischemia/reperfusion group (group B) and shenfu injection treatment group(group C), In group A, the rats received sham operation only. In group B, the animals recieved posterior limb ischemia 4 hours and reperfusion 4 hours. In group C, shenfu injection 2.0 mL/kg were injected iv 5 rain before ischemia and 15 min before reperfusion. The level of malondialdehyde (MDA), myeloperoxidase (MPO), wet weight/dry weight (W/D), cytochrome oxidase (CCO) and Ca^2+-ATPase in the skeletal muscle were measured at the end of the experiment. Shenfu injection can reduce the heightened level of MDA, MPO, W/D and protect the activity of CCO, Ca^2+-ATPase in the skeletal muscle, lighten the injury of the ultrastructure. Condition Shenfu Injection can protect ischemia/reperfusion injury in skeletal muscle.
