血宝含药血清和再障患者血清对骨髓基质细胞的影响 被引量:1

Effects of serum containing Xuebao and serum from patients with aplastic anemia on bone marrow mesenchymal cells

摘要 目的:探讨血宝含药血清和再生障碍性贫血(再障)患者血清对骨髓基质细胞的影响。方法:提纯的再障患者血清抑制蛋白组分加到骨髓基质细胞培养体系48h后,采用免疫组化法观察骨髓基质细胞形态学变化和TUNEL法测定骨髓基质细胞的凋亡。大鼠胃饲血宝7d后制成含药血清按比例加入培养液,培养骨髓基质细胞,观察细胞形态和通过计数了解血宝对骨髓基质细胞的刺激作用。结果:20例正常人骨髓基质细胞分别与0.1、1、10!g/mL的再障血清抑制蛋白组分共同培养后,Fas表达阳性率分别为(10.7±4.2)%(P>0.05)、(16.9±9.8)%(P>0.05)、(49.4±10.5)%(P<0.01),而对照组为(14.4±7.8)%;凋亡细胞发生率分别为(5.6±1.6)%(P>0.05)、(24.8±8.2)%(P<0.01)、(43.3±9.8)%(P<0.01),对照组为(4.6±1.5)%。血宝高、低剂量含药血清培养骨髓基质细胞5d后细胞计数分别为(5.0±0.5)×104(P<0.01)和(2.3±0.6)×104(P<0.05),空白对照组为(1.3±0.2)×104。结论:再障患者血清抑制蛋白组分与正常人骨髓基质细胞的凋亡有一定的量效关系。推测Fas-FasL系统可能是启动骨髓基质细胞凋亡的一条重要途径,血宝具有促进骨髓基质细胞增殖的作用。 Objective To investigate the effects of serum containing Xuebao and serum from patients with aplastic anemia on bone marrow mesenchymal cells. Methods Purified serum inhibitors from patients with aplastic anemia were cultured with normal bone marrow mesenchymal cells for 48 hours. The morphological changes of mesenchymal cells were observed using immunohistochemistry and apoptosis of the cells was detected by TUNEL. The serum containing Xuebao from rats gastrically administered with the medication for 7 days was cultured with the mesenchymal cells. Cell morphology was observed and the stimulative action of Xuebao on the cells was determined. Results The rate of Fas expression in the mesenchymal cells was ( 10.7 ± 4.2)% for 0.1 μg/mL of serum inhibitor cultivation (P 〉 0.05), ( 16.9 ± 9.8)% for 1 μg/ mL (P 〉 0.05), and (49.4 ± 10.5)% for 10 μg/mL (P 〈 0.01 ), as compared with (14.4 ± 7.8)% in the control group. The apoptosis rate was (5.6 ± 1.6)% (P 〉 0.05), (24.8_± 8.2)% (P 〈 0.01), and (43.3 ± 9.8)% (P 〈 0.01), respectively, while that was (4.6 ± 1.5)% in the controls. The count of mesenehymal cells was (5.0 ± 0.5)~104 umler cultivation for 5 days with serum containing Xuebao at a high dose (P 〈 0.01 ) or (2.3 ± 0.6)×10^4 under that with serum containing Xuebao at a low dose (P 〈 0.05), as compared to ( 1.3 ± 0.2)× 10^4 in the blank group. Conclusion There is some dose-effect relationship between serum inhibitor protein and apoptosis of normal bone marrow mesenchymal eells, suggesting that Fas-FasL system may be an important pathway triggering the cell apoptosis. Xuebao capsule plays a role in the proliferation of bone man'ow mesenchymal cells.
