摘要 目的:观察熊果酸(ursolicacid,UA)对胃癌细胞株MGC-803的抑制增殖作用。方法:培养胃癌细胞株MGC-803,以MTT比色法及克隆形成实验测定UA对胃癌细胞株MGC-803抑制增殖的作用。免疫印记法测定p-ERK1、cyclinD1及p21waf表达。结果:MTT实验及克隆分析法均显示UA明显抑制MGC-803细胞增殖,免疫/cip1/2印记法显示UA抑制p-ERK1、cyclinD1表达,同时上调p21waf/cip1/2表达。结论:UA可以抑制胃癌细胞株MGC-803增殖,其机制与影响p-ERK1及下游cyclinD1、p21waf/cip1/2表达有关。 Objective To observe the effects of ursolic acid (UA) on the proliferation of human gastric carcinoma cell line MGC-803. Methods Human gastric carcinoma cell line MGC-803 was cultured and the effects of UA on the proliferation of MGC-803 cells were determined by MTF assay and clone formation experiment. The expressions of p-ERK1/2, Cyclin D1 ,and p21^waf/eipl were detected by Western blot. Results The proliferation of MGC-803 cells was obviously suppressed, The expressions of p-ERK1/2 and cyclin D1 were inhibited whereas p21^waf/eipl expression was upregulated by UA. Conclusion UA can suppress the proliferation of gastric carcinoma cell line MGC-803, which is involved in the regulation of p-ERK1/2, cyclin D1, and p21^waf/eipl.
