摘要 目的观察Pokemon在肝细胞癌(HCC)、肝细胞癌远处转移、肝硬化患者和健康人群外周血中的表达特征及其影响因素。分析Pokemon作为HCC诊断、预测转移复发标记物的价值。方法研究对象分4组(n=40)。HCC合并乙型肝炎后肝硬化行肝移植患者、HCC远处转移合并乙型肝炎后肝硬化、乙型肝炎后肝硬化及健康人群各抽外周血2ml 1次。用实时定量逆转录-聚合酶链反应(RT-PCR)法检测Pokemon mRNA的表达。结果(1)Pokemon在HCC、肝硬化和健康人群外周血中的表达差异无统计学意义(P>0.05)。(2)HCC远处转移组Pokemon表达量、表达比例明显高于其余各组(P<0.05)。(3)HCC外周血中Pokemon表达水平与性别、年龄、HBSAG、HBV-DNA、AST、ALT、AFP无关(P>0.05)。结论HCC患者外周血中的Pokemon表达水平不高;肝硬化、健康人群组也可能存在Pokemon的表达。HCC远处转移组Pokemon表达水平明显高于HCC肝移植、肝硬化和健康人群组。监测外周血中Pokemon表达水平可能有助于诊断和预测HCC远处转移。 Objective The study was to use the quantitative RT-PCR (real time reverse transcfiptase polymerase chain reation, RT-PCR) to quantify the expression level of Pokemon gene in blood of HCC (hepatoceUular carcinoma, HCC ), HCC with distant place metastasis, hepatocirrhosis and normal crowd and to find out the differences, so that we could ensure which factors can affect expression level of pokemon in blood and evaluate the value of pokemon as a HCC marker. Methods There were 4 groups (n = 40) which included HCC, HCC with distant place metastasis, hepatocirrhosis and normal crowd group. Samples were drawed-off 2 ml blood. The expressions of Pokemon in serum were detected by RT- PCR. Results The expression level of Pokemon in HCC was not significantly different from hepatocirrhosis and normal crowd group. The expression level and ratio of Pokemon in HCC with distant place metastasis group was significantly higher than that in other groups. Conclusion The expression level of Pokemon in blood with HCC is not higher than that in hepatocirrhosis and normal crowd who maybe exist the expression of Pokemon. The expression level of Pokemon in HCC with distant place metastasis group is higher. To monitor the expression level of Pokemon in blood maybe useful for diagnosing and predicting HCC with distant place metastasis.
