红芪多糖HPS-3体外诱导人胃癌MGC-803细胞凋亡研究 被引量:11

摘要 目的:研究红芪多糖均一组分HPS-3体外对人胃腺癌MGC-803细胞增殖及凋亡的作用。方法:应用MTT法、显微荧光术和流式细胞术测定HPS-3对MGC-803细胞的增殖、细胞周期和诱导凋亡的影响,应用流式细胞术测定对Bc1-2和Bax蛋白表达的影响。结果:HPS-3对MGC-803细胞具有抑制增殖、G2/M期阻滞和诱导凋亡作用;HPS-3降低bcl-2蛋白表达,对Bax蛋白表达无明显作用。结论:HPS-3可通过抑制人胃腺癌MGC-803细胞的生长增殖、G2/M期阻滞、诱导细胞凋亡,降低bcl-2蛋白等作用机制发挥肿瘤作用。 Object: To study the effect of Hedysari polysaccharide HPS - 3 on the proliferation and apoptosis of human gastric cancer cell line MGC -803 in vitro. Methods: MTT assay,flow cytometry and fluorescence microscopy were used to examine the effects of HPS -3 in anti - proliferation, cell cycle arresting and apoptosis induction. Flow cytometry were used to detect the effects on protein expression of Bcl -2 and Bax. Results: HPS -3 could cause apparent inhibition on cell growth, induce G2/M phase arrest and apoptosis on MGC -803 cells. HPS - 3 could down- regulate bcl- 2 protein expression, but Bax protein expression showed unchanged. Conclusion: HPS -3 shows its anti - tumor effects by way of inhibiting the cell growth, arresting the G2/M phase, inducing apoptosis, down -regulating bcl -2 protein expression.
