川芎嗪抗大鼠免疫损伤性肝纤维化作用及机制研究 被引量:14

摘要 目的:观察川芎嗪(TMP)对大鼠免疫损伤性肝纤维化的治疗作用并探讨其相关机制。方法:建立猪血清诱导的大鼠免疫性肝纤维化模型后给予TMP干预,分别观察TMP对受试动物肝组织病理学分级、血清生化和血清纤维化指标的影响,免疫组化法检测TGFβ-1、TIMP-1在肝组织的阳性表达。结果:TMP 10、20和40mg/kg均可显著改善大鼠肝纤维化分级程度;显著降低大鼠血清ALT、AST、ALP水平,提高A/G比值;下调TGFβ-1、TIMP-1表达;TMP各剂量可不同程度的降低肝纤维化大鼠血清HA、LN、C-IV和PⅢNP水平。结论:TMP对大鼠实验性肝纤维化具有明显的治疗作用;其机制可能与抑制细胞因子TGFβ-1、胶原酶抑制因子TIMP-1表达增高有关。 Objective: To investigate the effects of tetramethylpyrazine (TMP) for chronicity liver fibrosis in rats and the possible mechanism . Methods: A model of liver fibrosis was set up by intraperitoneal injection porcine serum into rats. The effect of TMP( 10,20 and 40mg/kg) in the treatment of liver injury was evaluated by the degree of liver fibrosis, the serum biochemical indicator included ALT, AST and ALP, and fibrosis indicator included serum HA, LN,C-Ⅳ and PⅢNP. The expression of transforming growth factor-beta 1 (TGF-β1), tissue inhibitor of metalloproteinase-1 ( TIMP-1 ) in hver tissue were detected by immunohistochemical techniques. Results: The degree of liver fibrosis indexes were improved significantly; the serum ALT, AST, ALP level were depressed but the level of A/G were enhanced; the expression of TGF-β1, TIMP-1 were reduced by TMP. The serum HA, LN, C-Ⅳ and PⅢNP level in model rats were decreased by TMP. Conclusion: TMP has treament actions to rats with immunodamaged hepatic fibrosis. The mechanism of its effect is related with its inhibition the expression of TGF-β1 and TIMP-1 in rats liver.
