摘要
目的探讨牙髓慢失活剂小鼠口服半数致死量,为临床治疗提供安全、可靠的临床数据。方法取SPF级NIH种小鼠,同一性别每10只一组,试验前禁食14小时,正常饮水,雌雄各一组按最大灌胃容量40ml/kg的剂量给予上述供试液,一日给药1次,给药后连续观察7天,记录动物的反应、体重变化及死亡情况。结果小鼠灌胃给予的牙髓慢失活剂后雌、雄小鼠反应情况基本一样。低剂量组动物均出现兴奋、出汗,随着剂量的增加,动物均出现惊厥、角弓反张、死亡。结论NIH种小鼠灌胃给予牙髓失活剂的半数致死量LD50,雌性小鼠为916mg/kg,雄性小鼠为853mg/kg,以成人体重为60kg计算,即相当于成人临床用药量的20mg/kg的458倍和426倍。
Objective Dental pulp slow deactivator is the main agent for root channel therapy. The present study is to observe acute toxic reaction and lethal situation of mice induced by dental pulp slow deactivator with intragastric administration one time. Methods SPF grade NIH mice were divided into 10 mice per group at the same sex. Abrosia for 14 hours and normal hydroposia before experiment, dental pulp slow deactivator was given by max intragastric administration volume 40 ml/kg dose, one time per day. Reaction, change of body weight and lethal situation of mice were observed for 7 days. Results Reaction of mice given by dental pulp slow deactivator were uniformity at the different sex. Excitation and perspiration appeared in low dose group mice. As increasing of dose, convulsion and opisthotonos began to appear. Conclusions Median lethal dose of NIH mice given by dental pulp slow deactivator are 916 mg/kg in female mice and 853 mg/kg in male mice, which is equal to 458 times and 426 times of adult clinical drug consumption, if adult body weight is 60 kg.
