摘要
目的利用聚酰胺柱层析分离蜂胶中总黄酮,并运用紫外分光光度法(UV)对其进行含量测定。方法利用聚酰胺柱层析,干法上样,分别用30%、95%不同浓度的乙醇进行洗脱,盐酸—镁粉反应定性检测及分光光度法定量测定,并对分光光度法进行了改进。结果经定性检测,30%乙醇洗脱液几无黄酮,紫外分光光度法测量95%洗脱液中总黄酮的含量为0.956mg/ml。结论利用聚酰胺柱层析初步纯化蜂胶中总黄酮,并提供了测量蜂胶中总黄酮含量的一种快速、准确的方法—紫外分光光度法,为进一步分离其中的单体黄酮及研究其药理活性打下了基础。
Objective To separate the flavonoids in proplis by polyamide column chromatography and to establish the method of UV for determination of flavonoid. Methods Flavonoid of proplis was separated by polyamide column chromatography with 30%, 95% ethanol and determined by Hd- Mg reaction and ultraviolet- visible spectrophotometry. Results There was almost no flavonoid in eluate of 30% ethanol, the concentration of eluate of 95% ethanol was 0.956mg/ml by U- V Spectrophotometry. Conclusion The flavonoid was primarily purified by polyamide column chromatography, and a fast, accurate method-ultraviolet-visible spectrophotometry was put forword.
