摘要
目的研究一种绿僵菌固态发酵产壳聚糖酶的新方法。方法使用正交实验方法研究以日本根霉液态发酵后的菌丝体粉末为氮源,麸皮为碳源时,绿僵菌固态发酵产壳聚糖酶的最佳条件。结果在起始pH 5.0,27℃下培养120 h,c/N为1:4、固液比为1:1.4,绿僵菌发酵产壳聚糖酶活性最高。在优化的条件下,固态发酵产壳聚糖酶的最大活性可达35.08 U/g干培养基。
Objective To develop a new method for chitosanase production by Metarhizium anisopliae with solid - state fermentation. Methods The solid - state fermentation medium and technological conditions of Metarhizium anisopliae were optimized by orthogonal experiments. Conclusion The maximum activity of the chitosanase in the process of solid - state fermentation could Obtain in such conditions: an initial pHS.0, 27℃, the ratio of carbon source and nitrogen source was 1:4, the ratio of solid and liquid was 1 : 1.4, and the culture time was 120 hours. The best yield of chitosanase which could obtain in optimal conditions was 35.08U/g.
