摘要
应用MS+2.0mg·L(-1)2,4-D+0.5mg·:L(-1)BA作为培养基,附加500mg·L(-1)水解酪蛋白和w=3%蔗糖,可以从玉米叶片愈伤组织中直接诱导出体细胞胚胎.观察表明,玉米胚性细胞同非胚性细胞差别显著.玉米幼叶体细胞胚胎可能起源于单细胞;发育早期的胚性细胞团与周围薄壁细胞存在着明显的界限,推测可能同体细胞胚胎发生初期的生理隔离有关,它随着胚胎发育而逐渐消失.玉米体细胞胚胎胚柄发达,由一列或多列细胞组成.玉米叶片体细胞胚胎主要发生于愈伤组织的表皮层或表皮层下部.
Somatic embryos could be induced directlyfrom the most basalsegments of the inneryoung leaves of maize seedling (12-15d, 15-20cm tall) on the MS medium with 2.0 mg' L-12, 4-D and 0.5mg' L-1 BA, adding 500mg' L-1 CH and w=3% sucrose.
