摘要
目的研究三七总皂苷对严重烫伤大鼠心脏收缩舒张功能及心肌细胞凋亡发生的影响。方法通过大鼠40%体表皮肤Ⅲ度烫伤模型(时相点为伤后6,12,24h),应用生理记录仪监测、心肌组织切片脱氧核糖核酸末端转移酶介导的缺口末端标记(TUNEL)、染色等技术观察大鼠严重体表烫伤后心脏收缩舒张功能变化及心肌细胞凋亡情况,观察三七总皂苷对它们的影响,同时检测三七总皂苷对烫伤大鼠心肌组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量变化的影响。结果大鼠严重烫伤后心脏收缩舒张功能显著下降,心肌组织TUNEL阳性细胞数增加,心肌组织发生了脂质过氧化反应,烧伤后12h最显著。应用三七总皂苷后心脏功能下降趋势缓解、TUNEL阳性细胞率下降、心肌脂质过氧化反应减轻。结论烧伤后心肌组织发生了以心功能下降、心肌细胞凋亡为特征的心肌损害,三七总皂苷能减轻大鼠烧伤后的心肌功能损害,其机制与减轻脂质过氧化反应及降低心肌细胞凋亡发生率有关。
Objective To study the effect of panax notoginseng saponins on systolic, diastolic pressure and cell apoptosis of cardiac muscle in severely scalded rat. Methods 40% skin- full-thickness scald model was produced, the observing time was 6, 12, 24 h after scald, The function of heart was watched with physiological recorder. The apoptosis rate of heart was detected with the technology of terminal deoxynucleotidyl transferase TdT-mediated biotin dUTP nick end labeling (TUNEL). The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) were detected also. In the meantime, the effect of panax notoginseng saponins on the heart of scald rat was observed too. Results After scald, the function of heart decreased rapidly, the number of positive cells by TUNEL staining increased, lipid peroxidation response occurred. All these responses were significant at 12 h after scald. After panax notoginseng saponins was used, the decreasing tendency of heart function was slowed, the TUNEL positive cell rate decreased, lipid peroxidation response was lightened. Conclusion The function of heart decreased and cell apoptosis occurred after rat was scalded. Panax notoginseng saponins can lighten the injury of heart of scalded rat, the mechnism of this protection effect may be correlated with the capacity of decreasing lipid peroxidation response of rat heart.
