摘要
目的探讨聚凝氨-微板法筛查不规则抗体的可行性。方法使用聚凝氨-微板法对弱阳性IgG抗D组和阴性对照组进行检测,通过人工及酶标仪判读,观察各组不同静置时间凝集、沉降强度和光密度值变化与差异。结果弱阳性组与阴性对照组凝集和沉降差异显著并在一定时间段内随静置时间延长而明显增强;实验结果可经酶标仪进行自动化判读和记录;聚凝氨-微板法对弱阳性IgG抗D抗体检测的灵敏性高于试管法。结论为聚凝氨-微板法批量筛查不规则抗体的方法学建立提供实验基础。
Objective To study the possibility of screening red cell irregular antibody by polyhrene-microplate method. Methods By detecting weak positive anti-D antiserm and negtive serm with polybrene-microplate method, the diversity of coagulation, sedimentation and optical density are observed With microplate spectrophotometers and macrography. Results The diversity of the coagulation and sedimentation is markedly enhanced as time being prolonged. The experimental result can be conducted by the automated interpretation and records. The polybrene-mlcroplate method is more sensitive than the tube methed for the detection of weak positive anti-D antiserm. Conclusion The experimental basis is provided for the establishment of polybrene-microplate methodology.
