摘要
目的探讨雷公藤多甙对大鼠原位肝移植免疫抑制的作用机理。方法将120只肝移植SD大鼠分为4组:对照组(A组),TⅡ组(B组),CsA组(C组),TⅡ+CsA组(D组),分别检测移植术后各组1、3、5、7、14d外周血IL-2R的含量和脾脏淋巴细胞的IL-2活性及肝细胞凋亡相关基因Bcl-2、FasL的蛋白表达。结果三组治疗组的IL-2活性和IL-2R含量均明显低于对照组,三组治疗组间的比较是D组低于B、C组,上述均有统计学意义(P<0.05)。三组治疗组的Bcl-2的蛋白表达低于对照组,FasL的表达高于对照组,而三组间的比较:D组低于C组,C组低于B组,上述均有统计学意义(P<0.05)。结论雷公藤多甙通过对IL-2、IL-2R和Bcl-2、FasL的影响来达到免疫抑制作用,如果联合CsA效果会更好。
Objective To investigate the mechanism of GTW on the immunosuppression of orthotopic rat liver transplantation.Methods 120 orthotopic liver transplantation SD rats were divided into 4 groups：control group（A）,TⅡ group（B）,CsA group（C）,TⅡ ＋CsA group（D）.Serum levels of IL-2R、activity of IL-2 in spleen lymph cells and express of Bcl-2、FasL（coherent gene of hepatocyte apotosis） were detected respectively on 1、3、5、 7、14d after operation.Results Serum levels of IL-2R and activity of IL-2 were detected lower in 3 treated groups than in control group.Among the 3 treated groups,lower in D than in B and C（P〈0.05）.The result also showed that express of Bcl-2 lower in 3 treated groups than in control group,however,express of FasL higher in B、C、D than in A（P〈0.05）.Conclusion GTW performs its function of immunosuppression on the liver transplantation through IL-2、IL-2R and Bcl-2、FasL.There is combined effect of CSA and GTW on the immuno-suppression of liver transplantation.
