摘要
目的:观察狼毒大戟根中的不同提取物组分和不同化学成分对耐药型和非耐药型强毒人型结核杆菌的体外抑菌效果。方法:采用不同极性的溶剂对狼毒大戟根进行提取,得到6个提取组分,对其中的醋酸乙酯部分进行了柱层析和制备性高效液相层析分离,分离鉴定出狼毒乙素、狼毒乙素-4-O-β-D-吡喃葡萄糖苷、大黄素甲醚、没食子酸等4个单体成分。采用改良7H10琼脂培养基动态观察6个组分和4个单体成分对耐药型和非耐药型结核杆菌的抑制作用。结果:4种单体成分对耐药型和非耐药型结核杆菌均有显著的抑制作用。其中,狼毒乙素的作用最显著;6个提取组分中。乙醚、醋酸乙酯提取物时非耐药型和耐药型结核杆菌亦有不同程度的抑制作用,醋酸乙酯提取物作用最强。结论:狼毒大戟根中存在着抗结核杆菌特别是抗耐药型结核杆菌的活性部位及有效成分。
Objective : To observe the inhibiting effects of different extractions and constituents of root of Euphorbia fischeriana on tuberculosis bacillus or drug-resisting tuberculosis bacillus in vitro. Method: Six extractions were isolated from the roots of Euphorbia fischeriana by systematic extraction according to their polarity. Four constituents were isolated by column chromatography and preparing HPLC from the ethyl acetate extraction and identified as 2,4 dihydroxy-6-methoxy-3-methylacetophenone, 2,4 dihydroxy-6-methoxy-3- methylacetophenone-4-O-β-D-glucopyranoside, physcion and gallic acid. Tuberculosis bacillus and drug-resisting tuberculosis bacillus were cultured in improved 7H10 agar medium by the two-fold dilution method for four weeks, and were observed all the time. Result: Four constituents have antibacterial activity on tuberculosis bacillus and drug-resisting tuberculosis bacillus ,2,4 dihydroxy-6-methoxy- 3-methylacetophenone was the most effective. Of six extractions tested, ethyl acetate and ethyl ether extracts had antibacterial activity on both tuberculosis bacillus and drug-resisting tuberculosis bacillus, but to different extents, ethyl acetate extraction was the most effective. Conclusion: The effective extracts and constituents to tuberculosis bacillus and drug-resisting tuberculosis bacillus were contained in roots of Euphorbia fischeriana.
