奶牛离体PBMC和体内免疫应答的个体差异及其机理的研究

摘要 检测15头奶牛离体外周血单核细胞(PBMC)和体内产生抗体免疫应答的结果表明,1.0μg/ml的PWM或SEB使PBMC增殖达最高值,但在分泌Ig中却表现出不同作用;还发现在群体内离体细胞增殖强度的变异程度(C.V≤12.5%)小于分泌的Ig量的变异程度(C.V=31.9%~62.2%,其中个别Ig量超出x±2SD)。15头奶牛在实验期间(180d)的12次血清样品中IgM、IgG1和IgG2水平变异程度(C.V≤10%)也小于它们个体之间的变异程度(C.V=11.8~15.8%)。在对卵清蛋白为载体的异硫氰酸荧光素(FITC)的特异性反应中,初次和再次反应的抗体滴度(IgG1和IgG2)呈明显相关(r=0.698和0.867,n=15;P<0.01);而个体之间抗体滴度的变异程度大(C.V=28.4%~36.7%),个别超出x±2SD。分析表明,在PBMC分泌Ig水平或抗体反应滴度变异的个体与PBMC增殖强度和血清Ig水平无关。这些说明在免疫功能正常并相近的同种动物群体内,仍存在一定数量的个体。 This page reports the study of the immune response of boPBMC in vitro to mitogens in the proliferation or Ig production and of cattle in vivo to FITC with ovalbumin as carrier at the same time.PWM or SEB,which induced boPBMC strongly to proliferate in the concentration of 1.0μg/ml,could induce cells to produce Ig or not.It is found that there was smaller different extent among the tested cattle in the intensity of cell proliferation(C.V≤12.5%,n=15) than in the production of Ig(C.V≥31.9%,even 62.2%) by PWM or SEB (0.01~1μg/ml) induced PBMC, in some individual the produced Ig would exceed x±2SD.The level of Ig classes and IgG subclasses in the serum kept relative steady not only in the individual (C.V≤10%,n=12),but also in the group of these animals (C.V=11.8~15.8%,n=15) in the tested period about 180 days.Meanwhile,after immunization of FITC ovalbumin,the titers of anti FITC antibody (including their Ig classes)in sera from these animals showed significant difference (C.V≥28.4%~36.7%),even some exceeded x±2SD,(p<0.05).In addition,there was a linear regression between the antibody titers in the primary and secondary reactions (r=0.698,IgG 1;0.867,IgG 2,n=15,p<0.01);nevertheless no regression could be found between the antibody titers in vivo or the produced Ig in vitro and Ig levels in sera or cell proliferation in vitro. All these results suggest that the homogeneous population,which have normal immune function,could show significant difference in the response to an identical antigen or mitogen because of the immune response gene.
