白念珠菌细胞壁不溶性β-葡聚糖升高小鼠血清IL-1β、TNF-α的研究 被引量:4

摘要 为探讨白念珠菌β-葡聚糖升高白细胞的机制,通过培养白念珠菌来获取不溶性β-葡聚糖(Candida albicansinsoluble β-glucan,CAIBG),并以rhG-CSF作为阳性对照,计数实验组及对照组动物外周静脉血白细胞总数,用酶联免疫吸附试验(ELISA)检测IL-1β、TNF-α,观测小鼠血清中IL-1β、TNF-α的变化,以及IL-1β、TNF-α与CAIBG诱导的末梢血白细胞计数之间的相关性。结果发现在实验组小鼠外周血白细胞,血清IL-1β、TNF-α较对照组均有明显升高(P<0.05)。结果表明白念珠菌不溶性β-葡聚糖可以通过升高小鼠血清IL-1β、TNF-α来刺激机体白细胞产生增加。 To investigate the mechanism of inoluble β- glycan from cell wall of Candida a lbicans (CAIBG)to raise the count of leukocytes in peripheral blood of mice, CAIBG extracted from the cultured C. albicans CJ strain was injected into mice in a dosage of 51.2 mg/10 ml/kg, and blood was drawn at different periods after injection. The total count of leukocytes in periph eral blood and the contents of IL-1β and TNF-α in serum were determined by routine method and ELISA. The changes of IL-1β and TNF - α as well as their relationships to the leukocyte count in peripheral blood induced by CAIBG were observed. It was found that in the experimental group of mice, the leukocyte count in peripheral blood and the levels of IL-1β and TNF-α were increased obviously in eompareson with those of the control groups(P〈0.05). It is evident that the insoluble β-glycan of C. albicans can increase the count of ceukocytes in peripheral blood through elevation of the serum level of IL-1β and TNF-α.
