摘要
目的建立肠出血性大肠埃希菌O157∶H7体外黏附HeLa细胞模型,为进一步研究其致病机制奠定基础。方法以细菌黏附HeLa细胞数量为指标,分别评价细菌数量、细菌与细胞孵育时间对细菌黏附数量的影响,得到最佳条件,最后肌动蛋白荧光染色(FAS)试验鉴定模型。结果加入细菌为1×105CFU,细菌与细胞孵育4h时,黏附细胞的数量最佳,FAS检测发现该条件下细菌能够引起细胞特异性黏附、擦拭(attaching&effacing,A/E)损伤,模型建立成功。结论成功建立O157∶H7体外黏附HeLa细胞模型,为进一步研究其致病机制提供了条件。
Objective To establish Enterohemorrhagic Escherichia coli (EHEC) O157:H7 attached HeLa cell model for further study on its pathogenic mechanism. Methods HeLa cells and Escherichia coli O157:H7 were incubated together in different condition (amount of bacteria, incubation time). In regard to the amount of bacteria attached, we decided the best condition for O157:H7 to infect HeLa cells and identified the characterized A/E ( attaching and effacing) lesion by FAS (fluorescence actin staining). Results The best condition is that 1 × 10^5 CFU O157:H7 and HeLa cells were incubated together for 4 h, under which the specific A/E lesion can be observed. Conclusion EHEC O157:H7 attached HeLa cell model has been successfully established.
