摘要
对极大螺旋藻(Spirulina maxima)用热水抽提、脱蛋白、醇析得粗多糖,再经DEAE-Sephadex A-25柱层析纯化得到2个组分的多糖精品(SPSⅠ和SPSⅡ),分别经Sephadex G-150柱层析鉴定为单一组分,它的紫外可见光谱只出现195.00nm的单峰。在体外设计产生超氧阴自由基(O_2^+)、脂质自由基(R·)和羟自由基(OH·)等3个体系,分别加入2个组分不同浓度的多糖,结果表明:极大螺旋藻多糖具有显著的清除OH·的活性(P<0.05),但对O_2^+没有显著作用(P>0.1);在低浓度条件下(2.5×10^(-4)g/L),它们可以清除R·(P<0.05),SPSⅠ比SPSⅡ的活性显著;在较高浓度下清除R·的活性有所降低(P>0.2)。说明极大螺旋藻多糖的显著抗氧化作用主要表现在清除活性氧中最活泼的OH·方面。
The crude polysaccharide was extracted from Spindina maxima with hot water, and precipitated by ethanol after deproteinization. Two portions of refined polysaccharides (SPS Ⅰ and SPS Ⅱ) were prepared after further purification on DEAE-Sephadex A-25 column chromatography, and their homogeneity was examined with Sephadex G-150 column chromatography. The ultraviolet spectra showed their characteristic absorption at 195.00 run. In order to estimate the antioxidation activity of SPS Ⅰ and SPS Ⅱ , three systems of generating superoxide radical (O2T ), lipid radical (R ·) and hydroxyl radical (OH2T) respectively in vitro were designed. The results showed that both SPS Ⅰ and SPSⅡ from S. maxima had significant capacity of scavenging O2T (P <0.05), but no effect on O2T (P > 0.1); and that SPS Ⅰ could scavenge R · under lower concentration of polysaccharides (P < 0.05), while the capacity of scavenging R · of both SPS Ⅰ and SPS Ⅱ decreased in higher concentration (P > 0.2). These results demonstrated that the significant antioxidation activity of polysaccharides from S. maxima, was focused on scavenging OH· , the most highly reactive one of the oxygen radicals.
