Establishment and maintenance of longan embryogenic cell lines

摘要 本研究比较了生长调节剂、AgNO3、活性炭、碳源、基本培养基、光照度、胚发育时期以及基因型等对龙眼幼胚培养诱导愈伤组织的影响.从幼胚培养中,诱导出多种类型的愈伤组织,经继代培养并筛选,获得松散、生长旺盛、胚胎发生能力极强的松散型胚性愈伤组织.该类型胚性愈伤组织适宜建立悬浮细胞系和分离原生质体,并且转移至含体积分数为0.05椰子汁的MS固体培养基上,形成大量的胚状体.采用在附加1mgL-12,4-D的MS培养基与附加1mgL-12,4-D、0.5mgL-1KT和5mgL-1AgNO3的MS培养基交替继代培养。 The comparison of the effects of plant growth regulators, AgNO 3, active carbon, carbon sources, basal media, illuminance, embryo developmental stages and genotypes on the induction of calli from longan immature embryos were carried out, and several types of calli were induced in this study. Then, the friable and vigorous calli with strong embryogenetic capacity were selected in the process of subcultures. The selected calli were suitable for establishment of embryogenic suspensions and isolation of protoplasts, and a large number of cotyledonary embryoids formed after they were transferred into solid MS medium containing 0.05 volume fraction of coconut water. The embryogenic calli could be maintained for a long period when they were alternately cultured on the MS medium supplemented with 1 mg·L -1 2, 4 D and the other MS medium containing 1 mg·L -1 2,4 D, 0.5 mg·L -1 KT and 5 mg·L -1 AgNO 3.
