应用流式细胞术检测10种细胞株癌胚抗原表达的比较分析

摘要 目的为了解10种细胞株的癌胚抗原(carcinoembryonic antigen,CEA)表达,以便选择合适的靶细胞用于进一步研究。方法用抗CEA全人单克隆抗体C2-45(IgG1)以流式细胞术检测分析了10种细胞株。结果MKN-45、CHO-CEA、QGP-1和KATOⅢ为CEA阳性表达的细胞株,KNS-62和Hela-CEA为CEA表达稍弱的细胞株,HT-29和CaCO2为CEA微量表达的细胞株,CHO-MK1和MKN-74为CEA不表达的细胞株。大部分细胞株的CEA表达结果与文献报告基本一致,部分细胞株的CEA表达与文献不尽相同。结论CEA表达特性的改变有可能影响细胞株体外生物学行为从而导致实验结果的偏差,本实验结果可作为肿瘤细胞免疫研究重要的参考指标。 Objective To investigate the expression of carcinoembryonic antigen (CEA) in 10 cell lines for the further research. Methods Ten cell lines were detected and analyzed by flow cytometry using CEA specific human monoclonal antibody C2-45 ( IgG1 ). Results All of MKN-45,CHO-CEA,QGP-1 and KATO Ⅲ were obviously CEA-positive cell lines. The expression of CEA in KNS-62 and Hela-CEA were weaker than that in the four cell lines above. There was very low expression of CEA in HT-29 and CaCO2. There was almost no expression of CEA was detected in CHO-MK1 and MKN-74. The expressions of most CEA cell lines were almost coincident with these reported in other research. Conclusion The characteristic such as the expression of CEA lost in cell line may influence its biological behaviour and further lead to the uncertainty of the experiment.
