乌头碱水解转化规律的研究 被引量:40

摘要 目的:了解乌头碱的水解转化规律。方法:将乌头碱单体置纯水中,密封加热8,12,16,20,24 h,水解液碱化萃取,采用HPLC-MS分析主要水解产物;进一步采用中性氧化铝柱色谱分离,得到主要水解产物,并鉴定结构。结果:经HPLC-MS分析,鉴定主要水解成分峰为苯甲酰乌头原碱和乌头原碱。水解20 h,苯甲酰乌头原碱的转化率最高。结论:双酯型的乌头碱可水解转化为单酯型乌头碱和乌头原碱;在控制条件下,苯甲酰乌头原碱为主要水解产物。 Objective:To find out hydrolysis regularity of aconitine.Method: Aconitine was air-tighted and hydrolyzed in water for 8,12,16,20,24 h.The hydrolysis products were analyzed by HPLC-MS.A major hydrolysis product was isolated by alumina oxide column chromatography,and identified by ^1H and ^13C-NMR spectra.Result: HPLC-MS analysis shows that the major hydrolysis products are benzoylaconine and aconine.The hydrolysis can mostly be completed within 20 hours.Conclution: Diester aconitum alkaloid can be changed to monoester aconitum and aconine alkaloids.Under the controled condition benzoylaconine is a major hydrolysis products.
