摘要
重组肿瘤坏死因子对SW-480结肠癌细胞杀伤作用低敏感,但对细胞克隆生长的抑制作用很经。将TNF与5-Fu联合应用,单用TNF94u/ml深度对SW-480只有克隆生长抑制作用其杀伤率不到2%,5-Fu12.5mg/L浓度单用杀伤率只8.6%,但合有香TNF杀伤率提高1270倍,5-Fu刹伤率提高15倍,MMC0.625mg/L浓度单用杀伤率23.8%,单用TNF1.
rhTNF's killing effect on human colon carcinoma cell line SW-480 is low sensitive,whereas itscolony inhibiting effect is strong. The administration of rhTNF with concentration of 94 U/ml had an inhibiting effect on SW-480 with killing rate less than 2%. The administration of 5-Fu at 12. 5 μg/ml showed a killing rate of 8.6%. However, the effect of rhRNF rose 1270 times and 5-Fu 15 times when the two were used concomitantly. Furthermore, when used alone at 0.625 mg/mland 1.2×104U/ml,MMC's and rhTNF's killing rate was 23.8% and less than 5% , respectively. The evidents suggested that rhTNF plus 5-Fu or MMC produced obvious synergic effects in vitro. For clinical application, these synergic effects could reduce the dose of drugs, the toxic effects and the cost of therapy.
