整合素αvβ3对胶质瘤细胞增殖和侵袭力影响的实验研究 被引量:8

摘要 目的:研究整合素αvβ3对胶质瘤细胞增殖和侵袭能力的影响。方法:采用MTT比色法和增殖细胞核抗原(PCNA)免疫组化链霉亲和素-生物素-过氧化物酶复合物(SABC)法,观察不同浓度的整合素αvβ3抗体对体外培养的C6胶质瘤细胞增殖的影响;建立Boyden小室细胞侵袭模型,观察不同浓度的整合素αvβ3抗体对体外培养的C6胶质瘤细胞侵袭能力的影响。结果:对照组A值(0.2135±0.0325)、肿瘤细胞增殖抑制率(0)分别与实验组A值(0.1687±0.0403、0.1194±0.0512和0.0823±0.0338)和肿瘤细胞增殖抑制率[(31.56±3.36)%、(45.21±5.32)%和(68.27±4.49)%]比较,差异有统计学意义,F值分别为14.75和19.46,P均<0.01。实验组组间比较亦差异有统计学意义,F=7.86,P<0.05;F=13.77,P<0.01。对照组(0.7223±0.0318)与实验组(0.4592±0.0436,0.2967±0.0625)以及实验组组间PCNA表达水平的比较,差异有统计学意义,F=16.82,P<0.01;F=8.95,P<0.05。对照组(41.36±5.87)和实验组(32.53±3.89、18.82±4.02和13.71±2.94)以及实验组组间C6胶质瘤细胞透膜细胞计数比较,差异有统计学意义,F值分别为20.34和13.23,P均<0.01。结论:整合素αvβ3对胶质瘤的细胞增殖和侵袭性生长具有促进作用,其为抗整合素αvβ3治疗脑胶质瘤提供了实验依据。 OBJECTIVE: To investigate the effect of integrin αvβ3 on cell proliferation and invasive ability of C6 glioma cells in vitro. METHODS: The effect of various concentration of integrin αvβ3 monoclonal antibody on C6 glioma cells proliferation in vitro was detected by MTT method and the strepavidin biotin complex (SABC) immunohistochemistry technique. The effect of various concentrations of integrin αvβ3 monoclonal antibody on C6 glioma cells invasive ability in vitro was measured by Boyden chamber inva sive models. RESULTS: The differences between the A values (0. 2135±0. 0325) in control group and the A values (0. 168 7±0.040 3, 0.119 4±0.051 2, 0.082 3±0.033 8) in experimental groups, and the differences between the inhibited rates of tumor cell proliferation (0) in control group and the inhibited rates of tumor cell proliferation [(31.56±3.36)%,(45.21±5.32)%, (68.27±4. 49)%] in experimental groups, were significant (F=14.75,P〈0.01;F=19. 46,P〈0.01), and the differences of the A values and the the inhibited rates of tumor cell proliferation among the experimental groups were also significant (F= 7.86,P〈0.05;F= 13.77, P〈0.01). The expression level of proliferating cell nuclear antigen (PCNA) (0. 722 3±0. 031 8) in the control group was different significantly from those (0. 459 2±0. 043 6,0. 296 7±0. 062 5) in the experimental groups (F= 16.82,P〈0. 01), and the differences of the expression level of PCNA among the the experimental groups were significant,F= 8.95, P〈.0.05. The differences of the values of C6 glioma cells penetrating polyethylene perephthalate membrane between the control group (41.36 ± 5.87) and the experimental groups (32. 53±3.89, 18.82±4.02,13.71±2. 94) and among the the experimental groups were significant (F= 20. 34, P〈0.01; F= 13.23, P〈0.01). CONCLUSION: Integrin αvβ3 can positively regulate the cell proliferation and invasive ablity of glioma, and provide more experimental bases for glioma treatment by anti-αvβ3 integrin.
