RAPD标记法鉴定中药黄精及长梗黄精的研究 被引量:12

摘要 目的对中药黄精及主要掺伪品长梗黄精进行鉴别,确保中药使用的安全有效。方法采用原植物鉴定、性状鉴定、显微鉴定和RAPD手段,对随机引物进行筛选,通过1.8%琼脂糖凝胶电泳分离PCR产物。结果通过筛选23种RAPD引物,得到CTGGGGCTGA,GCTGGCTGAC,TCCCGAACCG三种引物,经PCR扩增产生足够的多态性谱带用于分析。结论RAPD技术为黄精与长梗黄精的区分提供了分子鉴别依据。 Objective For the safe use of Chinese traditional drug, Polygonatum sibiricum and its fake called P. filipes, were identified by a new method. Methods Character identification, microscopic identification and RAPD were used. All of the samples were amplified by PCR system and separated by 1.8% electrophoresis. Results 23 arbitrary premiers were evaluated for the analysis of RAPD the primers such as CTGGGGCTGA, GCTGGCTGAC ,TCCCGAACCG could yield enough polymorphic bands for the analysis. Conclusion The method RAPD can be used for the identification of P. sibiricum and P. filipes.
