芪术合剂对博莱霉素致大鼠肺纤维化白介素-8和转化生长因子-β_1的影响 被引量:1

The Effect of Qizhuheji on Interleu Kin-8 and Transforming Growth Factor-β_1 Expression in Lung Tissue of Pulmonary Fibrosis Rat Models

摘要 目的探讨中药芪术合剂对博莱霉素致大鼠肺纤维化的影响及其作用机制。方法将60只健康SD大鼠随机分为正常组、模型组、芪术合剂组和强的松组,经气管滴入博莱霉素复制肺纤维化大鼠模型,并分组予以治疗,于治疗后第3、7、21日分批处死大鼠,每组每次6只;用免疫组化法观察肺组织转化生长因子-β1的表达,并测定支气管肺泡灌洗液中中性粒细胞和白介素-8的水平。结果在第3、7、21日模型组IL-8和TGF-β1表达均较正常对照组明显增高,芪术合剂组与同期模型组比较明显降低,芪术合剂第3日组高于同期强的松组,而第7、21日则明显低于强的松组。结论早期应用芪术合剂可抑制IL-8和TGF-β1的产生,从而预防和阻止实验性大鼠肺纤维化的形成。 Objective: To investigate the effect and the mechanism of Chinese herbs, Qizhuheji(QZHJ) on bleomycin - induced pulmonary fibrosis in rats. Methods: 60 SD rats were divided into 4 groups randomly, the nomal group, the model group(treated with bleomycin), QZHJ group (treated with QZHJ), and the hydrocortisone group (trented with hydrocortisone) . Rats were sacrificed in batch on the 3rd, 7th and 21st day after treatment, then the concentration of interleukin - 8(IL -8) in bronchoalveolar lavage fluid (BALF) and the content of transforming growth factor - β1 (TGF - β1 ) of lung tissue were deected, while the severity of alveolitis and fibrosis were measured. Results: The concentration of TGF - β1 of lung tissue in the model group was higher than that in the normal group on the 3rd, the 7th and the 21st day, and it was lower in QZHJ group than that in the model group respectively. It was higher than that in the hydrocortisone group on the 3rd day, but lower on the other days. The changqing of severity of alveolitis and fibrosis and the content of IL- 8 in BALF in QZHJ were similar with that of TGF - β1. Conclusion: Early intervention with QZHJ could inhibit the secretion of IL - 8 and TGF - β1 and prevent the development of alveolitis and fibrosis of lungs in bleomycin - induced pulmonary fibrosis rats.
