摘要
目的:研究黄连与生地黄不同比例配伍对小檗碱在大鼠体内药动学的影响,探寻二者最佳配伍比例。方法:给各组大鼠分别灌胃给予黄连:生地黄为1:0,1:1,1:4,1:8水煎剂,采用高效液相色谱(HPLC)法测定大鼠体内小檗碱的血药浓度,血药浓度.时间数据经DAS2.0药代动力学软件分析,比较各组大鼠体内小檗碱药代动力学参数。结果:小檗碱在大鼠体内的代谢过程符合一级二室模型,权重形W=1/C^2,方差分析结果表明,4组中相同剂量的黄连配伍不同比例的生地黄,小檗碱Cmax,AUC0-t,AUC0-∞有显著差异(P〈0.05),其中,黄连与生地黄1:8组Cmax,AUC0-t,AUC0-∞最大。结论:黄连配伍大剂量的生地黄,可显著提高黄连中有效成分小檗碱的生物利用度。
Objective: To study the effect of Rhizoma Coptidis and Radix Rehmanniae with the different ratio on the pharmacokinetics of berberine in rats. Method : 24 rats were grouped to 4 groups randomly. Decoction, in which the proportion of Rhizoma Coptidis to Radix Rehmanniae is 1 : 0, 1 : 1, 1 : 4, 1 : 8 differently, was intragastrically given to the 4 groups. HPLC was used to determine concentration of berberine in serum. We adopted DAS 2.0 to analysis pharmacokinetic parameters of berberine. Result. The concen- tration-time curves was all fitted to two-compartment model with a weight of 1/C^2. Difference of 4groups in Cmax, AUC0-t, AUC0-∞ is significant ( P 〈 0. 05 ). Conclusion: Radix Rehmanniae of large dose can effectively enhance berberine' s bioavailability in rats.
