9L/Wistar大鼠脑胶质瘤动物模型的建立

摘要 目的建立稳定的9L/Wistar大鼠脑胶质瘤动物模型。方法24只近交系雄性Wistar大鼠,采用立体定向技术在大鼠右尾状核接种1×106个9L细胞,观察大鼠生存状态;接种后8天行MRI检查,观测颅内肿瘤生长情况。大鼠死亡后,取脑制作病理切片,行HE染色后观察肿瘤组织形态。结果大鼠接种肿瘤细胞30天内全部死亡,平均生存时间23.38±1.60天。脑内均见肿瘤形成,未见肿瘤颅外生长。肿瘤周边界线明显,未见包膜。瘤内新生血管丰富,可见出血坏死。结论该方法建立的大鼠脑胶质瘤动物模型稳定可靠,符合恶性胶质肉瘤生物学特性。该模型是理想的脑胶质瘤实验研究材料。 Objective The establishment stable big rat brain gum quality lump animal model of the 9L/Wistar. Methods 24s are near to hand over to fasten the male Wistar big rat, adopting a stereoscopic definite direction technique to inoculate 1 × 10^69L cells in the big rat fight tail form pit, observing great rat existence appear- ance;Go a MRI check for 8 days after inoculating, prognosticate tumor a growth a circumstance inside . The big rat takes the brain creation pathology slice after dying, observing tumor organization appearance after going HE dyeing. Results the big rat inoculate tumor cell all death in 30 days, equally existing time 23.38± 1.60 days. All see tumor formation inside brain, do not see tumor grow outside. Tumor week the boundary line is obvious, didn' t see a pack of film. The new - born blood vessel is abundant inside lump, it is thus clear that bleed bad dead. Conclusion the big rat brain gum quality lump animal of that method establishment model stability credibility, match malignant gum quality tumor biology characteristic. That model is an ideal brain gum quality lump experiment to study material.
