摘要
目的:为更好的进行钩端螺旋体病监测,为预防措施提供依据。方法:建立PCR检测方法,进行灵敏度、特异性研究。探索钩端螺旋体PCR扩增DNA条件。对浙江省2006年分离菌11株进行PCR鉴定,对龙游、常山捕获的一百只鼠和一百只蛙肝、脾、肾标本在提取其DNA后进行PCR检测。结果:所建立PCR检测方法具备较高灵敏度和较好特异性。对2006年分离的钩体菌株进行的PCR鉴定结果与血清学鉴定结果相符。鼠和蛙肝、脾、肾标本进行PCR检测,PCR检测阳性率10%,钩体菌株培养分离率仅为1%。2χ=15.58,P<0.05有显著差异。测序及培养均能证实PCR检测结果具有特异性。结论:钩体PCR检测方法的建立能更加准确的反映野生动物自然带毒率。钩体PCR检测方法可推广应用。
Objective: To strengthen the surveillance and provides the basis for the preventive strategies of leptospirosis. Methods:PCR was chosen to detect leptospira. The sensitivity and specificity of the assay was evaluated. Optimize the amplified condition of PCR and apply it to the detection of 11 isolates of Zhejiang Province in 2006. 200 renal, splenic and hepatic specimens from rats and frogs were detected with the assay after the extraction of their genome. Results: The approach we have developed has high sensitivity and specificity. PCR results on leptospira isolates of Zhejiang Province in 2006 were in accordance with that of serological detection. By using PCR to detect the plucks of rats and frogs, the positive rate was 10% while the seperation rate was only 1% by using traditional culture method. Significant differences were found between them(χ^2 = 15.58, P 〈 0. 05 ). Conclusion :The developed method for detecting leptospira with PCR is useful. It can reflect the natural virulent rate of leptospira in wild animals more accurate and supply evidence for correct diagnosis and treatment of leptospirosis. We should popularize and use this assay in the detection of leptospira.
