重度苯中毒患者T细胞受体Vβ亚家族TRECs的分析 被引量:1

摘要 目的检测重度苯中毒患者外周血23个TCRVβ-Dβ1TRECs的存在情况,进一步了解重度苯中毒患者胸腺近期各Vβ亚家族初始T细胞的输出情况。方法分别在5×104和1×104个外周血单个核细胞水平下,采用半巢式对12例重度苯中毒患者和10例正常人的23个TCRVβ-Dβ1TRECs进行扩增。结果在5×104和1×104细胞水平下,苯中毒组检出各TCRVβ-Dβ1TRECs数均显著低于对照组(P<0.05)。在5×104细胞水平时,对照组检测到23个TCRVβ-Dβ1TRECs,苯中毒组仅检测到17个TCRVβ-Dβ1TRECs,其中12个TCRVβ-Dβ1TRECs检出率与对照组相比明显降低(P<0.05)。结论本研究提供了苯中毒患者外周血中各TCRVβ-Dβ1TRECs存在情况。苯中毒患者TCRVβ-Dβ1TRECs检出数量和检出率低于正常人,提示重度苯中毒患者胸腺近期输出功能存在部分缺陷和低下的情况。 Objective To detect the existence of T-cell receptor (TCR) Vβ-Dβ1 TRECs in peripheral blood mononuclear cells in patients with severe benzene poisoning and to understand the recent thymic output of naive T cells of different TCRVβ subfamilys. Methods 23 TCRVβ-Dβ1 TRECs were amplified by semi-nested PCR in DNA from peripheral blood mononuclear cells of 12 patients with severe benzene poisoning and 10 normal individuals at 5 × 10^4 and 1 × 10^4 cell concentration, respectively. Results The numbers of detectable TCRVβ-Dβ1 TRECs in the patients were significantly lower than those in control groups at 5 × 10^4 and 1 ×10^4 cell concentration, respectively. At 5 × 10^4 cell concentration, all of TCRVβ-Dβ1TRECs were detected in con- trol groups and only 17 TCRVβ-Dβ1TRECs were detected in the poisoned groups, the rate of TCRVβ-Dβ1 TRECs in the poisoned groups were significantly lower than those in control groups. Conclusion This is the first report of the detection of TCRVβ- Dβ1 TRECs in patients with severe benzene poisoning. A fewer numbers and lower rate of TCRVβ-Dβ1 TRECs in the patients suggested that patients with severe benzene poisoning had partial deficient function and lowered recent output of their thymus.
