摘要
目的探讨苯乙酸对大肠癌细胞株HCT-8的诱导分化作用。方法应用[3H]-TdR掺入法与细胞计数法、流式细胞技术、电子显微镜检测PA对大肠癌细胞HCT-8增殖的抑制作用、细胞周期各时相及其超微结构的变化。结果HCT-8细胞增殖抑制率与PA剂量呈显著正相关关系;应用PA后,HCT-8细胞增殖抑制于G0/G1期,大量细胞堆积于S期末期,不能完成全部DNA合成,细胞核变规整,细胞质增多。结论PA对大肠癌HCT-8细胞存在时间和剂量依赖性增殖抑制作用,可提高大肠癌细胞HCT-8的分化程度,具有广阔的临床应用前景。
Objective To investigate the effect of Phenylacetate induced differentiation on colorectal cancer cell.Methods Proliferation inhibitory rate was evaluated by means of [3 H]-thymidine incorporation and cell count, cell cycle and super-microstructure were observed through flow cytornetry and electronic microscope. Results There was a close-dependent effect between the does of PA and proliferation inhibitory of HCT-8 cell, the percentage of HCT-8 cells in G0/G1 phase and G2 + M phase reduced significantdy and S phase increased relatively. Nucleus of cell changed from irregular shape to roundoid shape. Conclusion It has been demonserated that PA have a time and does-dependent growth ihibition effect and can increase the differentiation degree of colorectal tumor HCT-8 cells.
