HSV-TK/GCV系统对卵巢癌细胞OVCAR3杀伤作用的研究

摘要 目的评价HSV-TK/GCV系统对卵巢癌细胞OVCAR3的杀伤作用及旁观者效应。方法用含HSV-TK重组腺病毒载体转染人卵巢上皮性癌细胞OVCAR3,用四甲基偶氮唑蓝(MTT)法计算细胞存活率以评价杀伤作用;转染的OVCAR3细胞(TK+)与未转染的OVCAR3细胞(TK-)以不同比例混合,再加入GCV,MTT法计算细胞存活率以评价旁观者效应。结果HSV-TK/GCV系统可明显杀伤OVCAR3细胞,在感染复数(MO I)为100时,平均细胞存活率为26%;HSV-TK/GCV系统对该细胞的杀伤作用有明显的"旁观者效应",当TK+细胞占10%、20%、30%、40%、50%时,细胞平均存活率分别为82%、52%、41%、35%、32%。免疫细胞化学检测OVCAR3细胞的细胞膜上有Cx43的表达。结论HSV-TK/GCV系统对人卵巢上皮性癌细胞OVCAR3有明确的杀伤作用及旁观者效应。 Objective To study the killing action and the bystander effect of HSV - TK gene on ovarian cancer cells OVCAR3. Methods Cells of human ovarian cancer, OVCAR3 were transfected by adenovirus vector carrying HSV - TK gene. The effect of killing action and bystander effect were determined by the cells growth inhibitory rate with methyl thiazolyl tetrazolium. Results HSV - TK/ GCV system could kill OVCAR3 cells obviously. When the multiplicity of infection (MOI) was 100, the average survival rate was 26%. Bystander effect of HSV - TK/GCV system was obvious. When the ratio of TK^+ cells wasl0%, 20%, 30%, 40% and 50%, the average survival rate was 82%, 52%, 41%, 35% and 32%, respectively. Immunocytochemistry confirmed the expression of connexin 43 protein on the membrane of OVCAR3 cells. Conclusion The killing action and bystander effect of HSV - TK gene on ovarian cancer cells OVCAR3 are obvious.
