摘要
基于KIO4在H2SO4介质和沸水浴中可氧化偶氮氯膦Ⅲ褪色,而蛋白质能灵敏地阻抑该氧化褪色反应,建立了测定痕量蛋白质的新方法。蛋白质测定范围为0-6μg/mL,方法的检出限为2.9×10^-10g/L,回收率为96%-109%。测定了动力学参数,并对反应机理做了研究。该方法可用于实际样品的测定。
A new method for the detennination of trace bovine serum albumin (BSA) was proposed. It was based on the inhibitory effect of protein on discoloring reaction between KIO4 and chlomphosphonazo Ⅲ in dilute stulfuric acid solution at 100℃. The calibration graph is rectilinear from 0 to 6 μg/mL for BSA and the detection limit is 2.9 × 10^-10 g/L. Kinetic parameters and reaction mechanism have been studied. This method is simple and sensitive. It has been applied satisfactorily to the detennination of BSA in milk samples.
