红法夫酵母与环状芽孢杆菌混合培养破壁提取虾青素的研究 被引量:3

摘要 环状芽孢杆菌能以红法夫酵母细胞壁为唯一碳源进行发酵产胞壁溶解酶,将红法夫酵母与环状芽孢杆菌混合培养,可使环状芽孢杆菌在生长的同时产酶并逐渐溶解酵母细胞壁,从而提取虾青素。通过对红法夫酵母与环状芽孢杆菌两阶段混合培养条件的优化,总结出红法夫酵母产虾青素及酶法破壁提取的适宜工艺为:培养基初始糖浓度为30g/L,环状芽孢杆菌的最佳接种时间为红法夫酵母培养60~72h,接种量为10mL/L(1010个细胞/mL),接种后最适培养温度为30℃,pH为6.5,总发酵时间为120h,在此条件下,红法夫酵母虾青素产量可达到8960.2μg/L,虾青素最终提取率可达到96.8%。 Bacillus circulans could use the yeast cell wall as the sole carbon source to produce lytic enzyme. The yeast-wall was hydrolyzed and astaxanthin could be extracted from Phaffia rhodozyma by the mixed culture of Phaffia rhodozyma and Bacillus circulans. In this paper, results of the two-stage batch mixed cultures of Phaffia rhodozyma ATCC 66270 and Bacillus circulans A1. 383-2 indicated that the suitable incubation time and glucose concentration for Phaffia rhodozyma cultivation in the first stage were 60h~72h and 30g/L. During the second stage of the mixed culture, 10 mL/L(10^10 cells/mL)inoculum of Bacillus circulans A1. 383-2 supported the highest extractability of astaxanthin. The optimal temperature and pH for the astaxanthin extraction in the mixed culture were 30℃ and pH 6.5, respectively, and the total fermentation time was 120h. Under these conditions, the final astaxanthin production of Phaffia rhodozyma could reach 8 960.2μg/L and over 96.8% of the astaxanthin could be extracted during the second stage of this twostage mixed fermentation.
