摘要
目的:制备抗淋球菌外膜蛋白单克隆抗体,建立淋病快速诊断方法。方法:采用乙酸锂提取淋球菌WⅠ群和WⅡ/WⅢ群外膜蛋白,以此为抗原免疫BALB/C小鼠,融合、筛选抗淋球菌外膜蛋白杂交瘤细胞。结果:筛选出抗WⅠ群外膜蛋白杂交瘤2株,分泌的单克隆抗体类别为IgG1;抗WⅡ/WⅢ外膜蛋白杂交瘤2株,分泌的单克隆抗体类别为IgG2a和IgG2b。腹水单克隆抗体效价达107以上。4株单克隆抗体与脑膜炎球菌等细菌无交叉反应。对56例临床标本采用夹心ELISA法进行检测,结果发现敏感性达97%,特异性达100%。结论:该单克隆抗体可以用于淋病的快速诊断。
Objective: Produce monoclonal antibodies against outer membrane protein of Neisseria gonorrhoeae and set up methods of rapidity diagnosis. Methods: The outer membrane protein of Neisseria gonorrhoeae WⅠ and WⅡ/WⅢ were extracted with lithium acetate buffer. Results: We got 4 strains hybrid cell, which of 2 hybrid cells producing monoclonal antibodies against outer membrane protein of WⅠ secreted IgG 1, and the other 2 hybrid cells producing monoclonal antibodies against outer membrane protein of WⅡ/WⅢ secreted IgG 2a and IgG 2b . The titers of antibodies in ascites were more than 10 7 non-cross reactions were found with the strains of N. meningitides etc. The ELISA test was evaluated against the culture method of N. gonorrhoeae. The clinical specimens were collected from 56 swabs. The ELISA had a sensitivity of 97% and specificity of 100% were compared with the culture method. Conclution: The monoclonal antibodies could be used for rapidity diagnosis of gonorrhea.
